When using intercalating dyes, we suggest using the original cleaning reagent. The provided default passivation buffers (BSA + Pluronics) have also been replaced by Casein, a single step passivation buffer. It is still possible to select BSA + Pluronics as an option. —

The Buffer solutions and beads ‘Basics’ kit includes all the standard materials for your optical tweezers experiments. The materials are estimated for 3 months of C-Trap use.

The ‘Basics’ kit contains the following:

  • Phosphate-buffered saline (PBS) based running buffer 10x
  • Cleaning reagents*
  • Passivation buffer: choose between casein (recommended) or BSA + Pluronics*
  • Streptavidin or Anti-digoxigenin coated beads of different sizes

The kit contains polystyrene beads to attain the best trapping conditions depending on your scientific queries and experimental designs. While large diameter beads are easier to trap and are suitable for long DNA tethers (> 3000 bp), small diameter beads are particularly advantageous for short DNA tethers (< 3000 bp) and to measure protein unfolding and conformational changes at the nanoscale.

Specifications

All variations of the kit contain:

  • Running buffer 10x: 30 ml, 3 containers | (typical dilution factor 1:10). PBS with 50 mM sodium azide and 5 mM EDTA. Store at room temperature.

When selecting Casein passivation buffer (the recommended option), the kit contains:

  • Casein: 3 ml | 1% (typical dilution factor 1:20). Store at -20°C.
  • Alkaline cleaning reagent 50x (recommended for default cleaning): 3 ml, 4 containers (typical dilution factor 1:50). Store at room temperature.
  • Cleaning reagent (only recommended when using fluorescent intercalators): 300 ml. Contains bleach. Store at room temperature.
  • Reducing agent: 15 ml | (typical dilution factor 1:30). Contains sodium thiosulfate. Store at room temperature.

When selecting BSA/Pluronics passivation buffer, the kit contains:

  • Passivation buffer BSA: 4 containers, 4 ml | 1% (Typical dilution factor 1:10). Store at -20°C.
  • Passivation buffer Pluronic: 15 ml | 5% (Typical dilution factor 1:10). Store at room temperature.
  • Cleaning reagent: 300 ml. Contains bleach. Store at room temperature.
  • Reducing agent: 15 ml | (typical dilution factor 1:30). Contains sodium thiosulfate. Store at room temperature.

The kit contains a variable choice of beads:

  • Streptavidin-coated polystyrene beads (⌀ 0.7–0.9 μm / ⌀ 1.0–1.4 μm / ⌀ 1.5–1.9 μm): 60 μl | 1% (w/v) (typical dilution factor 1:1000). Beads in PBS with 3 mM sodium azide, with specific diameter (e.g., 0.8 μm) within the given range. Volume-based on the single selection of bead size. Store at +4°C.
  • Streptavidin-coated polystyrene beads (⌀ 4.0–4.9 μm): 600 μl | 0.5% (w/v) (typical dilution factor 1:100). Beads in PBS with 3 mM sodium azide, with specific diameter (e.g., 4.5 μm) within the given range. Volume-based on the single selection of bead size. Store at +4°C.
  • Anti-digoxigenin-coated polystyrene beads (⌀ 0.7–0.9 μm / ⌀ 2.0–2.4 μm): 600 μl | 0.1% (w/v) (typical dilution factor 1:100). Beads in PBS with 3 mM sodium azide, with specific diameter (e.g., 0.8 μm) within the given range. Volume-based on the single selection of bead size. Store at +4°C.
