The hairpin labeling and tethering kit provides a highly efficient system to characterize DNA/RNA structures using high-resolution single-molecule optical tweezers. Short handles will result in a higher force resolution, while long handles are better suitable for experimental workflows involving fluorescence. 

The labeling procedure relies on a DNA/RNA hairpin ligation to the functionalized DNA handles. The hairpin of interest (provided by the user) requires a flanking 15 nt DNA sequence both at the 5’ end and the 3’ end.

The kit contains a control hairpin containing the flanking 15 nt DNA sequences that can be used as quality control for the labeling and tethering procedures.

Specifications

  • Biotinylated (1045 bp or 2512 bp) and digoxigenin-labeled (1052 bp or 2528 bp) DNA handles:

85 μl. Handles mix (50/50) with overhangs that anneal with the hairpin’s flanking sequences. Store at -20 °C.

  • DNA Ligase:

10 μl. Store at -20 °C.

  • 10x Ligase buffer: 

25 μl | 10x. Store at -20 °C.

  • Control DNA

10 μl | 5 μM. DNA hairpin that can anneal with the handles’ overhangs that can be used as a quality control. Store at -20 °C.

  • Streptavidin coated polystyrene beads (1.0-1.4 µm): 

12 μl | 1% (w/v). Beads in PBS with 3 mM sodium azide, with a specific diameter (e.g 1.36 μm) within the given range. Store at +4 °C.

  • Anti-digoxigenin-coated polystyrene beads (0.7-0.9 µm):

60 μl | 0.1% (w/v). Beads in PBS with 3 mM sodium azide, with a specific diameter (e.g. 0.86 μm) within the given range. Store at +4 °C.
