The hairpin labeling and tethering kit provides a highly efficient system to characterize DNA/RNA structures using high-resolution single-molecule optical tweezers. Short handles will result in a higher force resolution, while long handles are better suitable for experimental workflows involving fluorescence.
The labeling procedure relies on a DNA/RNA hairpin ligation to the functionalized DNA handles. The hairpin of interest (provided by the user) requires a flanking 15 nt DNA sequence both at the 5’ end and the 3’ end.
The kit contains a control hairpin containing the flanking 15 nt DNA sequences that can be used as quality control for the labeling and tethering procedures. For the sequence of the control hairpin, have a look at the tab “Product resources”.
