Easier, more reproducible data – starting with the LUMICKS protocols

Protocol

Cleaning and passivation protocol (recommended)

When using any of the standard reusable LUMICKS flow cells, it is important to follow a robust cleaning protocol with alkaline cleaning reagent between experiments to avoid cross-contamination. After cleaning, the fluidics can be passivated with a casein to prevent non-specific interactions of your proteins with the fluidics walls.

Nucleosome microscale assembly protocol

Using the Nucleosome tethering kit, nucleosomes can be tethered in optical tweezers to study how DNA binding proteins and nucleosomes interact and regulate each other.

Generating single-stranded DNA

The single-stranded DNA is shipped as double-stranded DNA that is biotinylated at the 5’ and 5’ ends of the same strand. After melting the DNA one of the strands will fall off and a single stand remains.
