The DNA structural mechanics kit (for training) is the standard kit for your first optical tweezers experiments. This kit is perfect for calibrating your optical- tweezers equipment and training new users. You can use fluorescent intercalators (SYTOX Orange, YOYO-1, or DiTO-3) to visualize force-dependent structural changes on a DNA molecule (for more information on biotinylated double-stranded DNA with 2xATTO 647N included in the kit, see this page. Kits ordered before July 2023 contain DNA without ATTO647N fluorophores). Their specificity to double-stranded DNA enables to distinguish these regions from single-stranded DNA regions that are formed upon DNA denaturation induced by stretching the molecule.

Specifications

  • Biotinylated double-stranded DNA (48,502 bp) with 2x ATTO 647N: 20 μl | 20 ng/μl. Bacteriophage λ-DNA in TE buffer (10 mM Tris HCl pH 8.0 and 1 mM EDTA). Store at +4°C
  • SYTOX Orange / YOYO-1 / DiTO-3: 5 μl | 5 mM / 1 mM / 5 mM. Choose between SYTOX Orange (preferred option, green laser required), YOYO-1 (blue laser required), and DiTO-3 (choose when only red laser present). Store at -20°C
  • Streptavidin-coated polystyrene beads (⌀ 4.0–4.9 μm): 200 μl | 0.5% (w/v). Beads in PBS with 3 mM sodium azide, with a specific diameter (e.g., 4.5 μm) within the given range. Store at +4°C
