The DNA–protein interaction kit (for training) can kick-start your single-molecule experiments, allowing you to study the kinetics of enzymatic activities associated with DNA replication. The biotinylated pKYB1 DNA (8,398 bp) contains a 25-nucleotide long 5′-overhang that serves as a loading site for DNA polymerases (for more information on the DNA molecule, see this page). This kit is ideal for extracting real-time data of DNA replication processes, including polymerization dynamics, proofreading activities, and pause states.

T7 DNA polymerase:

20 μl, 200 Units | 10,000 U/ml. Store at -20°C.

dNTP mix:

300 μl | 10 mM of each nucleotide. Store at -20°C.

10x Polymerase buffer:

500 μl | 10x. Store at -20°C.

BSA passivation reagent:

2 ml | 1%. Store at -20°C.

Pluronic passivation reagent:

2 ml | 5%. Store at RT.

Biotinylated double-stranded DNA (8,398 bp) with overhang:

20 μl | 20 ng/μl. Linearized plasmid pKYB1 in TE buffer (10 mM Tris HCl pH 8.0 and 1 mM EDTA). Store at +4°C.

Streptavidin-coated polystyrene beads (⌀ 1.5–1.9 μm):

25 μl | 1% (w/v). Beads in PBS with 3 mM sodium azide, with a specific diameter (e.g., 1.7 μm) within the given range. Store at +4°C.
