Product Description

The standard-assay kit for your C-Trap™ instrument. This kit allows you to perform a standard experiment in which DNA polymerase activity can be measured. Contains the following:

  • T7 DNA polymerase
  • Biotinylated ds pKYB1 DNA with a 25-nt 5′-end overhang
  • Casein reagent
  • Streptavidin coated polystyrene beads (1.76 µm)
  • Streptavidin coated polystyrene beads (4.5 µm)
  • Immersion oil
  • Phosphate-buffered saline (PBS) based buffer
  • u-Flux™ syringes

Additional Information

T7 DNA polymerase

200 µl, 400 Units – 2000 U/ml (typical dilution factor 1:500) T7 DNA polymerase with 8x Histidine tag. In 50% glycerol. Can be supplied with E.coli thioredoxin for increased processivity.

Biotinylated ds pKYB1 DNA

25 µl – 20 ng/µl (typical dilution factor 1:1000) double-stranded expression vector pKYB1 (8393 bp) with a 25-nt 5′-end overhang. Biotin-labeled on the 5′ and 3′ ends of one of the strands, diluted in TE buffer (10 nM Tris HCl pH 8.0 and 1 nM EDTA).

Casein reagent

50 ml – 1% (w/v) casein reagent in PBS buffer. For passivation of u-Flux™ microfluidics system.

Polystyrene beads (1.76 µm)

500 µl – 5% (w/v) (typical dilution factor 1:500) streptavidin coated polystyrene microspheres with a diameter of 1.76 µm in water. Binds to biotinylated DNA (e.g. the provided pkYB1) and can be optically manipulated using the C-Trap™.

Polystyrene beads (4.5 µm)

1000 µl – 5% (w/v) (typical dilution factor 1:100) streptavidin coated polystyrene microspheres with a diameter of 4.5 µm in water. Binds to biotinylated DNA (e.g. the provided pkYB1) and can be optically manipulated using the C-Trap™.

Immersion oil

20 ml – Non-fluorescence immersion oil for condensor immersion.

PBS based buffer

100 ml – Phosphate-buffered saline buffer with 5 mM sodium azide (NaN3) and 0.5 mM EDTA.

u-Flux™ syringes

30 pieces – 3 ml syringes that perfectly fit in the u-Flux™ system.
