This double-stranded pKYB1 DNA (8,398 bp) with a 25-nucleotide long 5′-overhang is biotinylated (2 biotins on 3’ end and 3 biotins on 5’ end) and nick-free, making it ideal for measuring DNA–protein interactions and enzymatic activities on DNA. The 5’ overhang serves as a loading site for enzymes (for example, T7 DNA polymerase) to study replication processes, including polymerization, proofreading, and replication pause states. The short length of the DNA also makes it ideal for measuring small structural changes of the DNA resulting from, for example, protein binding or buffer conditions.
